Product Overview
This rabbit polyclonal antibody targets death-associated protein 1 (gene DAP) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 11.2 kDa, and the supplier reports an observed band at about 15 kDa.
Death-associated protein 1 (DAP1) is a small, conserved proline-rich protein. It was first identified in screens for genes needed for interferon-γ-induced cell death, and has since been linked to repression of autophagy downstream of mTOR and, more recently, to ribosome hibernation, where it binds eIF5A near the ribosome exit tunnel to keep ribosomes inactive and stable.

Samples with a confirmed band (WB): human U-937 cells, human RT4 cells, human PC-3 cells, human HeLa cells, human HepG2 cells, human U2OS cells, human HL-60 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): SiHa cells.
Samples with a confirmed signal (flow cytometry): HL-60 cells.
Immunogen
Synthetic peptide from the C-terminal region of human DAP1/DAP. Sequence identity with the mouse and rat orthologs: 92.9%.
Reactivity Notes
The supplier lists reactivity with human. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
