Product Overview

Clone 27D63 is a rabbit monoclonal antibody that recognizes DDB1- and CUL4-associated factor 7 (gene DCAF7) in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 38.9 kDa, and the supplier reports an observed band at about 39 kDa.

DCAF7 (also known as WDR68) is a WD40-repeat scaffold protein that associates with the kinases DYRK1A and HIPK2 and helps regulate their activity and localization. Work in animal models shows it is needed for craniofacial development, and it also influences GLI1-dependent transcription. DCAF7 is studied in developmental signaling and kinase regulation.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human HepG2 cells, human MCF-7 cells, rat C6 cells, mouse RAW264.7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human DCAF7.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places DDB1- and CUL4-associated factor 7 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
