Product Overview
This rabbit polyclonal antibody targets ATP-dependent RNA helicase DDX1 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry, ELISA and immunoprecipitation. The predicted molecular weight is 82.4 kDa, and the supplier reports an observed band at about 88–90 kDa.
DDX1 is an ATP-dependent RNA helicase of the DEAD-box family that can unwind RNA-RNA and RNA-DNA duplexes. It takes part in several RNA processes, including tRNA splicing as part of the tRNA ligase complex, and has been linked to RNA clearance at DNA double-strand breaks. DDX1 is studied in RNA metabolism, DNA repair and viral replication.

Samples with a confirmed band (WB): human K562 cells, human Caco-2 cells, human SH-SY5Y cells, human RT4 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat testis tissue, mouse testis tissue, mouse ovary tissue, human colon tissue, mouse brain tissue, rat brain tissue, human colon cancer tissue, human lung cancer tissue, human ovarian cancer tissue, human pancreas cancer tissue, human skin cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ATP-dependent RNA helicase DDX1 in the nucleus, cytoplasm, cytoplasmic granule and mitochondrion. Tissue expression noted by UniProt (human): Highest levels of transcription in 2 retinoblastoma cell lines and in tissues of neuroectodermal origin including the retina, brain, and spinal cord. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
