Product Overview

Anti-Dengue virus type 1 prM Polyclonal Antibody is an affinity-purified rabbit polyclonal antibody raised against the pr region of the prM (precursor membrane) protein of Dengue virus type 1 (DENV-1), polyprotein residues Gly122–Arg205. It is supplied unconjugated in liquid form and is validated by the manufacturer for ELISA, immunohistochemistry and Western blot.

prM is one of the three structural proteins cleaved from the dengue virus genome polyprotein, together with the capsid protein C and the envelope protein E. In the immature virion prM forms heterodimers with E, and its N-terminal pr segment covers the E fusion loop, preventing premature fusion while the particle transits the secretory pathway. During maturation the host protease furin cleaves prM into pr and M in the acidic trans-Golgi network; pr stays associated with E until the particle is secreted and dissociates at neutral extracellular pH, while M remains in the mature virion. Because cleavage is often incomplete, prM-containing particles circulate in infected hosts, and prM is a recognised target of the human antibody response to dengue. Antibodies to prM are therefore used to detect immature or partially mature virus, to study virion maturation, and in serological and antibody-response research.

Immunogen & Antigen

The antibody was raised against an E. coli-derived recombinant fragment of the DENV-1 genome polyprotein spanning Gly122–Arg205, then purified on an antigen affinity column. In the UniProt reference entry P17763 the prM chain occupies residues 115–280, of which the pr peptide is 115–205 and the M protein is 206–280. The immunogen therefore lies entirely within the pr segment of prM, ending at the furin cleavage site (Arg205). The antibody pool is enriched for epitopes in pr, and coverage of the mature M protein (residues 206–280) is not claimed.

The five accession numbers listed by the manufacturer (P17763, P27909, P33478, P27913, P27912) are all UniProt entries for the DENV-1 genome polyprotein from different isolates (Nauru/West Pac/1974, Brazil/97-11/1997, Singapore/S275/1990, Jamaica/CV1636/1977 and Thailand/AHF 82-80/1980); P27912 and P27913 are partial entries of about 790 residues, so residue numbering on this page follows the full-length entry P17763. The manufacturer does not state which isolate the immunogen sequence derives from.

Reactivity Notes

Manufacturer-stated reactivity is Dengue virus type 1. Behaviour against the other dengue serotypes (DENV-2, DENV-3, DENV-4) and related flaviviruses is not characterised by the manufacturer. Do not assume it in either direction: polyclonal antibodies raised against the pr region are commonly cross-reactive across dengue serotypes and related flaviviruses, so this product should not be treated as DENV-1-selective without in-house confirmation. The species-level reactivity value (Dengue virus) on this listing is a catalogue filter roll-up, not a manufacturer claim. The manufacturer offers separate anti-prM polyclonal antibodies for DENV-2, DENV-3 and DENV-4.

Because the immunogen lies within the pr segment, signal is expected from prM-containing (immature or partially mature) material rather than from fully processed M. Include an uninfected control when staining infected cells or tissue.
