Product Overview
Clone 2B5 is a mouse monoclonal antibody that recognizes desmin in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 53.5 kDa, and the supplier reports an observed band at about 54 kDa.
Desmin (DES) is the main intermediate filament protein of skeletal, cardiac and smooth muscle. It links neighboring myofibrils at the Z-disks and connects them to the sarcolemma, nucleus and mitochondria, helping muscle cells resist mechanical stress. Desmin is a standard muscle marker, and mutations cause desmin-related myopathies and cardiomyopathies.

Samples with a confirmed band (WB): rat heart tissue, rat skeletal muscle tissue, mouse heart tissue, mouse skeletal muscle tissue, human K562 cells, rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, human oesophagus squama cancer tissue, human skeletal muscle tissue, rat skeletal muscle tissue, mouse skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with a confirmed signal (flow cytometry): THP-1 cells.
Immunogen
Recombinant human Desmin fragment (Met1–Thr304), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 97%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places desmin in the cytoplasm, cell membrane, nucleus and cell tip. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
