Product Overview

Clone BDF-4 is a rabbit monoclonal antibody that recognizes desmin in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 53.5 kDa, and the supplier reports an observed band at about 54 kDa.

Desmin (DES) is the main intermediate filament protein of skeletal, cardiac and smooth muscle. It links neighboring myofibrils at the Z-disks and connects them to the sarcolemma, nucleus and mitochondria, helping muscle cells resist mechanical stress. Desmin is a standard muscle marker, and mutations cause desmin-related myopathies and cardiomyopathies.

Target Desmin (gene DES; UniProt P17661, human)

Validated Applications

Western blot 1:1000–1:5000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): rat heart tissue, rat skeletal muscle tissue, mouse heart tissue, mouse skeletal muscle tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1: 10000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human prostate tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human desmin.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places desmin in the cytoplasm, cell membrane, nucleus and cell tip. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on feline samples?
A.Reactivity with feline samples has not been tested by the supplier. Aligning the immunogen sequence with the feline ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 53.5 kDa; observed band at about 54 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human desmin.
