Product Overview
Clone 4E3 is a mouse monoclonal antibody that recognizes dihydroorotate dehydrogenase (gene DHODH) in human, mouse and rat samples and is validated for Western blot and ICC/IF. The predicted molecular weight is 42.9 kDa, and the supplier reports an observed band at about 43 kDa.
Dihydroorotate dehydrogenase (DHODH) is an enzyme of the inner mitochondrial membrane that oxidizes dihydroorotate to orotate using ubiquinone as the electron acceptor. This is the fourth step of de novo pyrimidine synthesis and links it to the respiratory chain. DHODH is the target of leflunomide and teriflunomide and is studied in nucleotide metabolism, immunology and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
Samples with a confirmed band (WB): human HeLa cells, human A431 cells, human HepG2 cells, human MCF-7 cells, rat testis tissue, rat liver tissue, mouse testis tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): MCF-7 cells.
Immunogen
Synthetic peptide from the N-terminal region of human DHODH. It differs from the mouse sequence at four residues and from the rat sequence at two residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places dihydroorotate dehydrogenase in the mitochondrion inner membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
