Product Overview
Clone 21D17 is a rabbit monoclonal antibody that recognizes dickkopf-related protein 3 (gene DKK3) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 38.4 kDa, and the supplier reports an observed band at about 50 kDa.
Dickkopf-3 (DKK3) is a secreted glycoprotein related to the Dickkopf family but divergent from DKK1, DKK2 and DKK4, and it does not clearly bind LRP5/6. It is often reduced in tumors and has been proposed as a tumor suppressor, while also being studied in kidney and cardiac fibrosis. Its precise mechanisms are still being characterized.

Samples with a confirmed band (WB): human U251 cells, human U2OS cells, rat thymus tissue, rat brain tissue, mouse thymus tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human liver cancer tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human dkk3.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes dickkopf-related protein 3 as a secreted protein. Tissue expression noted by UniProt (human): Highest expression in heart, brain, and spinal cord. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
