Key elements and design rationale

  • Target: DNMT1 (DNA (cytosine-5)-methyltransferase 1).
  • Antibody format: Monoclonal; clone AODB-4; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

DNMT1 (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Methylates CpG residues. Preferentially methylates hemimethylated DNA. Associates with DNA replication sites in S phase maintaining the methylation pattern in the newly synthesized strand, that is essential for epigenetic inheritance. Associates with chromatin during G2 and M phases to maintain DNA methylation independently of replication. It is responsible for maintaining methylation patterns established in development. DNA methylation is coordinated with methylation of histones. Mediates transcriptional repression by binding to HDAC2. In association with DNMT3B and via the recruitment of CTCFL/BORIS, involved in activation of BAG1 gene expression by modulating dimethylation of promoter histone H3 at H3K4 and H3K9. . Reported cellular localization context: Nucleus . Tissue expression notes (as provided): Ubiquitous; highly expressed in fetal tissues, heart, kidney, placenta, peripheral blood mononuclear cells, and expressed at lower levels in spleen, lung, brain, small intestine, colon, liver, and skeletal muscle. Isoform 2 is less expressed than isoform 1. .

Research relevance and current trends

  • Research context keywords from the source record include: 2339,Epigenetics and Nuclear Signaling.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.

Workflow ideas (metafield): Validate DNMT1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect DNMT1 expression by Western blot in cell or tissue lysates, Detect DNMT1 in FFPE tissue sections by immunohistochemistry, Localize DNMT1 by immunofluorescence/immunocytochemistry in cultured cells

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 200 kDa; calculated MW: 183165 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 200 kDa
  • Cellular localization (provided): Nucleus .
  • Tissue details (provided): Ubiquitous; highly expressed in fetal tissues, heart, kidney, placenta, peripheral blood mononuclear cells, and expressed at lower levels in spleen, lung, brain, small intestine, colon, liver, and skeletal muscle. Isoform 2 is less expressed than isoform 1. .

We ordered your anti-Dnmt1 Rabbit Monoclonal antibody for WB on cervix carcinoma erythroleukemia in a previous experiment. I am using rat, and We intend to use the antibody for IF next. I would like examining cervix carcinoma erythroleukemia as well as leukemic t-cell in our next experiment. Could give a recommendation on which antibody would work the best for IF?
I have checked the website and datasheets of our anti-Dnmt1 Rabbit Monoclonal antibody and it seems that M00172 has been validated on rat in both WB and IF. Thus M00172 should work for your application. Our Boster satisfaction guarantee will cover this product for IF in rat even if the specific tissue type has not been validated. We do have a comprehensive range of products for IF detection and you can check out our website bosterbio.com to find out more information about them.
My team were satisfied with the WB result of your anti-Dnmt1 Rabbit Monoclonal antibody. However we have seen positive staining in cervix carcinoma nucleus. using this antibody. Is that expected? Could you tell me where is DNMT1 supposed to be expressed?
From what I have seen in literature, cervix carcinoma does express DNMT1. Generally DNMT1 expresses in nucleus. Regarding which tissues have DNMT1 expression, here are a few articles citing expression in various tissues: Cervix carcinoma, Pubmed ID: 17081983, 18220336, 18669648, 20068231 Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163 Leukemic T-cell, Pubmed ID: 19690332 Liver, Pubmed ID: 24275569 Prostatic carcinoma, Pubmed ID: 15057824
