Product Overview

Clone 24T75 is a rabbit monoclonal antibody that recognizes transcription factor Dp-1 (gene TFDP1) in human, mouse and rat samples and is validated for flow cytometry, IHC and Western blot. The predicted molecular weight is 45.1 kDa, and the supplier reports an observed band at about 50 kDa.

DP-1 (TFDP1) is the main dimerization partner of E2F transcription factors. E2F–DP heterodimers bind E2F sites in promoters of genes needed for DNA replication and cell-cycle progression, and their activity is controlled by the retinoblastoma protein, so DP-1 is studied in cell-cycle regulation and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): human Raji cells, human A431 cells, human THP-1 cells, human 293T cells, rat PC-12 cells, rat C6 cells, mouse RAW264.7 cells, mouse Neuro-2a cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Samples with confirmed staining (IHC): human breast cancer tissue, human colorectal adenocarcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human DP1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places transcription factor Dp-1 in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Highest levels in muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Flow cytometry 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 45.1 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human DP1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
