Product Overview
This rabbit polyclonal antibody detects tumor necrosis factor receptor superfamily member 21 (gene TNFRSF21) in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 71.8 kDa, and the supplier reports an observed band at about 80–120 kDa.
Death receptor 6 (DR6, TNFRSF21) is a TNF receptor family member with a cytoplasmic death domain that can trigger apoptotic cell death. It restrains oligodendrocyte maturation and myelination, dampens T-cell and Th2 cytokine responses, limits antibody production, and has been linked to caspase-8-driven pyroptosis, so it is studied in neurobiology, immunology and cell death research.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human K562 cells, human Raji cells, human Sw620 cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse Raw1647 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human gastric cancer tissue, human bladder cancer tissue, human renal carcinoma tissue, human melanoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): A549 cells.
Immunogen
Synthetic peptide from the middle region of human DR6/TNFRSF21. Sequence identity with the mouse and rat orthologs: 84.2%.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places tumor necrosis factor receptor superfamily member 21 in the cell membrane. Tissue expression noted by UniProt (human): Detected in fetal spinal cord and in brain neurons, with higher levels in brain from Alzheimer disease patients (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
