Product Overview
This rabbit polyclonal antibody detects proheparin-binding EGF-like growth factor (gene HBEGF) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 23.1 kDa, and the supplier reports an observed band at about 23 kDa.
Heparin-binding EGF-like growth factor (HB-EGF, HBEGF) is an EGF family ligand made as a membrane-anchored precursor that is shed to release a soluble growth factor. It binds EGFR and ERBB4, and it is needed for normal heart valve formation. The membrane form is also the cellular receptor for diphtheria toxin. HB-EGF is studied in wound healing, heart development and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human Jurkat cells, human U251 cells, rat C6 cells, mouse lung tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung adenocarcinoma tissue, human appendiceal adenocarcinoma tissue, mouse lung tissue, rat respiratory smooth muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): human colorectal adenocarcinoma tissue.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Synthetic peptide from the middle region of human DTR/HBEGF.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
