Vendor description (summary): Boster Bio Anti-ECSIT Antibody Picoband® catalog # A07966-3.

Biological background

Biological context: RNA-binding protein, which may be required during spermatogenesis.

Expression and localization notes: cellular localization: Nucleus. Cytoplasm., tissue context: Mainly expressed in testis. Expressed to a lower level in thymus. Weakly or not expressed in heart, liver, brain, placenta, lung, skeletal muscle, kidney and pancreas..

Common research applications

  • Western blotting (WB): Compare ECSIT levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of ECSIT in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify ECSIT-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: Evolutionarily conserved signaling intermediate in Toll pathway, mitochondrial (ECSIT), also known as SITPEC, is a protein that in humans is encoded by the ECSIT gene. Activation of NF-kB as a result of Toll-like receptor (TLR) and IL-1 receptor signaling is a major component of innate immune responses. Signals from these receptors are relayed by a number of adapter molecules such as TRIF, TIRAP, and MyD88 to kinases such as IRAK and other intermediates such as TNF receptor associated factor (TRAF)-6. ECSIT (evolutionarily conserved signaling intermediate in Toll pathways) was initially identified as a cytoplasmic protein interacting specifically with TNF receptor associated factor (TRAF)-6 in the TLR pathway. Recently however, ECSIT has also been shown to be required for bone morphogenetic protein (Bmp) signaling and mesoderm formation during mouse embryogenesis, indicating the possibility of cross-talk between the TLR/IL-B and Bmp signaling pathways.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Nucleus. Cytoplasm.
  • Tissue details: Mainly expressed in testis. Expressed to a lower level in thymus. Weakly or not expressed in heart, liver, brain, placenta, lung, skeletal muscle, kidney and pancreas.
