Key elements and design rationale

  • Target: EDNRB (endothelin receptor type B); UniProt: P24530
  • Antibody format: Mouse, Monoclonal, clone Clone: 15C3, Mouse IgG2b
  • Molecular weight: 45 kDa
  • Applications: WB, IHC, Flow Cytometry, ELISA

Vendor description (summary): Boster Bio Anti-EDNRB Antibody Picoband® (monoclonal, 15C3) catalog # M01041-1.

Biological background

Biological context: Non-specific receptor for endothelin 1, 2, and 3. Mediates its action by association with G proteins that activate a phosphatidylinositol-calcium second messenger system.

Expression and localization notes: cellular localization: Cell membrane. Multi-pass membrane protein., tissue context: Expressed in placental stem villi vessels, but not in cultured placental villi smooth muscle cells..

Common research applications

  • Western blotting (WB): Compare EDNRB levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of EDNRB in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify EDNRB-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Endothelin receptor type B, also known as ETB is a protein that in humans is encoded by the EDNRB gene. The protein encoded by this gene is a G protein-coupled receptor which activates a phosphatidylinositol-calcium second messenger system. Its ligand, endothelin, consists of a family of three potent vasoactive peptides: ET1, ET2, and ET3. Studies suggest that the multigenic disorder, Hirschsprung disease type 2, is due to mutations in the endothelin receptor type B gene. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane. Multi-pass membrane protein.
  • Tissue details: Expressed in placental stem villi vessels, but not in cultured placental villi smooth muscle cells.
  • Research category: G Protein Signaling,Signal Transduction,Signaling Pathway

Would M01041-1 anti-EDNRB antibody (monoclonal, 15C3) work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
As indicated on the product datasheet, M01041-1 anti-EDNRB antibody (monoclonal, 15C3) as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
