Product Overview
Clone ECH-5 is a rabbit monoclonal antibody that recognizes early endosome antigen 1 (gene EEA1) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 162.5 kDa, and the supplier reports an observed band at about 162 kDa.
Early endosome antigen 1 (EEA1) is a long coiled-coil protein that binds phosphatidylinositol 3-phosphate and active RAB5 on early endosomes. It tethers incoming vesicles for fusion and is one of the most widely used early endosome markers in cell biology.

Samples with a confirmed band (WB): human HeLa cells, human 293T cells, rat brain tissue, rat C6 cells, mouse brain tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human EEA1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places early endosome antigen 1 in the early endosome membrane and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
