Product Overview
This rabbit polyclonal antibody targets epidermal growth factor receptor (gene EGFR) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 134.3 kDa, and the supplier reports an observed band at about 180 kDa.
Epidermal growth factor receptor (EGFR, also called ErbB1 or HER1) is a receptor tyrosine kinase activated by EGF, TGF-α and related ligands. Ligand binding triggers receptor dimerization and activation of RAS/MAPK, PI3K/AKT and other pathways that drive proliferation and survival, and EGFR is a major target in cancer research.

Samples with a confirmed band (WB): human placenta tissue, human U87 cells, human A431 cells, human A549 cells, human HeLa cells, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human mammary cancer tissue, mouse liver tissue, rat liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A431 cells.
Samples with confirmed staining (ICC/IF): human lung cancer tissue.
Samples with a confirmed signal (flow cytometry): A431 cells.
Immunogen
Recombinant human EGFR fragment (Asn36–Asn497), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places epidermal growth factor receptor in the cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane and nuclear membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
