Product Overview
This rabbit polyclonal antibody detects eukaryotic initiation factor 4A-I (gene EIF4A1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 46.2 kDa, and the supplier reports an observed band at about 46 kDa.
eIF4A1 is a DEAD-box RNA helicase and part of the cap-binding eIF4F complex. It unwinds secondary structure in mRNA 5′ untranslated regions so the small ribosomal subunit can bind and scan, and it is studied in translation control and as a target of natural product inhibitors such as silvestrol.

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human A431 cells, human HEK293 cells, human HepG2 cells, human A549 cells, human U87 cells, human K562 cells, rat PC-12 cells, mouse spleen tissue, mouse thymus tissue, mouse lung tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, mouse intestine tissue, human lung cancer tissue, rat intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HepG2 cells, U87 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places eukaryotic initiation factor 4A-I in the cytoplasm and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
