Product Overview
This rabbit polyclonal antibody targets receptor tyrosine-protein kinase erbB-4 (gene ERBB4) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 146.8 kDa, and the supplier reports an observed band at about 180 kDa.
HER4 (ERBB4) is an EGFR-family receptor tyrosine kinase activated by neuregulins and some EGF-family ligands. It is needed for normal development of the heart, nervous system and mammary gland, and its cleaved intracellular domain can move to the nucleus to regulate gene expression.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
Samples with a confirmed band (WB): human MCF-7 cells, human 293T cells, human HeLa cells, rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, mouse brain tissue, rat brain tissue, human mammary cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Synthetic peptide from the N-terminal region of human erbB 4. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places receptor tyrosine-protein kinase erbB-4 in the cell membrane. Tissue expression noted by UniProt (human): Expressed at highest levels in brain, heart, kidney, in addition to skeletal muscle, parathyroid, cerebellum, pituitary, spleen, testis and breast. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
