Product Overview

Clone BCI-5 is a rabbit monoclonal antibody that recognizes receptor tyrosine-protein kinase erbB-2 (gene ERBB2) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 137.9 kDa, and the supplier reports an observed band at about 185 kDa.

HER2 (ERBB2) is a receptor tyrosine kinase of the EGFR family that has no known ligand of its own but is the preferred partner for other ErbB receptors. Heterodimers containing HER2 signal strongly through MAPK and PI3K-AKT pathways. HER2 amplification drives a subset of breast and gastric cancers, making it one of the most studied oncology targets.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:20
Flow cytometry 1:20

Samples with confirmed staining (IHC): rat stomach tissue, human breast carcinoma tissue, human esophageal carcinoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (ICC/IF): SKBR cells.

Immunogen

Synthetic peptide based on the sequence of human erbB2 (HER2).

Reactivity Notes

The supplier lists reactivity with human. UniProt places receptor tyrosine-protein kinase erbB-2 in the cell membrane and cell projection. Tissue expression noted by UniProt (human): Expressed in a variety of tumor tissues including primary breast tumors and tumors from small bowel, esophagus, kidney and mouth. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 137.9 kDa; observed band at about 185 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human erbB2 (HER2).
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
