Product Overview
This rabbit polyclonal antibody detects complement factor D (gene Cfd) in mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 28.4 kDa, and the supplier reports an observed band at about 27 kDa.
Complement factor D (CFD, adipsin) is a serine protease that starts the alternative complement pathway by cleaving factor B bound to C3b. It is produced mainly by adipocytes and circulates in plasma, and it is studied in innate immunity, complement disorders and adipose tissue biology.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat RH35 cells, rat PC-12 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): mouse lung tissue, rat lung tissue, rat small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Recombinant rat Factor D fragment (Ile26–Ala263), expressed in E. coli. Sequence identity with the human and rat orthologs: 69% and 84.5%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt describes complement factor D as a secreted protein. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
