Product Overview
This rabbit polyclonal antibody detects complement factor H (gene Cfh) in mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 139.1 kDa, and the supplier reports an observed band at about 150 kDa.
Complement factor H (CFH) is a major plasma regulator of the alternative complement pathway. It binds C3b on host cells, speeds the decay of the C3 convertase and acts as a cofactor for factor I, protecting self tissues from complement attack; variants of CFH are well established in age-related macular degeneration and atypical hemolytic uremic syndrome.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Mouse) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Mouse) |
Samples with a confirmed band (WB): mouse liver tissue, rat liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): HEPA1–6 cells.
Immunogen
Recombinant mouse Factor H/Cfh fragment (Asn136–Thr447), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt describes complement factor H as a secreted protein. Tissue expression noted by UniProt (mouse): CFH is one of the most abundant complement components in blood where the liver is the major source of CFH protein in vivo. in addition, CFH is secreted by additional cell types including monocytes, fibroblasts, or endothelial cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
