Product Overview

This rabbit polyclonal antibody targets F-box only protein 32 (gene FBXO32) in human and mouse samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 41.6 kDa, and the supplier reports an observed band at about 42 kDa.

FBXO32, also called atrogin-1 or MAFbx, is a muscle-enriched F-box protein that serves as the substrate-recognition part of an SCF E3 ubiquitin ligase. It is strongly induced during skeletal muscle atrophy and targets proteins for proteasomal degradation, so it is widely used as a marker in muscle wasting and atrophy research.

Validated Applications

Western blot 0.25–0.5 µg/mL (Mouse)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): mouse heart tissue, mouse skeletal muscle tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): HeLa cells.

Samples with a confirmed signal (flow cytometry): PC-3 cells.

Immunogen

Recombinant human FBXO32 fragment (Gln6–Phe355), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places F-box only protein 32 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Specifically expressed in cardiac and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
