Product Overview
This rabbit polyclonal antibody recognizes amyloid beta precursor protein binding family B member 1 (gene APBB1) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 77.2 kDa, and the supplier reports an observed band at about 65 kDa.
APBB1 (Fe65) is an adaptor protein that binds the cytoplasmic domain of the amyloid precursor protein (APP). With the APP intracellular domain released by γ-secretase it forms a nuclear complex that regulates transcription, and it also takes part in the DNA damage response, so it is studied in Alzheimer's disease research.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue, HeLa cells, U87 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue, human glioma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of human FE65. It differs from the mouse sequence at two residues and from the rat sequence at three residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places amyloid beta precursor protein binding family B member 1 in the cell membrane, cytoplasm, nucleus and cell projection. Tissue expression noted by UniProt (human): Highly expressed in brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
