Product Overview
Clone 9D8 is a mouse monoclonal antibody that recognizes fumarate hydratase, mitochondrial (gene FH) in human, monkey, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 54.6 kDa, and the supplier reports an observed band at about 48 kDa.
Fumarate hydratase (FH, fumarase) is a tricarboxylic acid (TCA) cycle enzyme in mitochondria that converts fumarate to L-malate; a cytosolic form also exists. Loss of FH causes fumarate to build up, and inherited FH mutations cause hereditary leiomyomatosis and renal cell cancer, so FH is studied in cancer metabolism and mitochondrial function.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
Samples with a confirmed band (WB): K562 cells, human placenta tissue, COS-7 cells, HL-60 cells, Caco-2 cells, U2OS cells, A549 cells, rat thymus tissue, rat testicular tissue, rat stomach tissue, mouse testicular tissue, mouse kidney tissue, NIH3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human intestinal cancer tissue, human lung cancer tissue, rat liver tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of human FH. Sequence identity with the mouse and rat orthologs: 100% and 97.8%, respectively.
Reactivity Notes
The supplier lists reactivity with human, monkey, mouse and rat. Tissue expression noted by UniProt (human): Expressed in red blood cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
