Purification Immunogen affinity purified.
Reconstitution Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Cellular localization Cytoplasm . Nucleus . Shuttles between the cytoplasm and nucleus. Largely nuclear in unstimulated cells. In osteoblasts, colocalizes with ATF4 and RUNX2 in the nucleus (By similarity). Insulin-induced phosphorylation at Ser-256 by PKB/AKT1 leads, via stimulation of Thr-24 phosphorylation, to binding of 14-3-3 proteins and nuclear export to the cytoplasm where it is degraded by the ubiquitin-proteosomal pathway. Phosphorylation at Ser-249 by CDK1 disrupts binding of 14-3-3 proteins and promotes nuclear accumulation. Phosphorylation by NLK results in nuclear export. Translocates to the nucleus upon oxidative stress-induced phosphorylation at Ser-212 by STK4/MST1. SGK1-mediated phosphorylation also results in nuclear translocation. Retained in the nucleus under stress stimuli including oxidative stress, nutrient deprivation or nitric oxide. Retained in the nucleus on methylation. .
Concentration Adding 0.2 ml of distilled water will yield a concentration of 500 μg/ml.

Product Overview

This rabbit polyclonal antibody targets forkhead box protein O1 (gene FOXO1) in human and rat samples and is validated for Western blot. The predicted molecular weight is 69.7 kDa, and the supplier reports an observed band at about 70 kDa.

FOXO1 is a forkhead transcription factor and a main target of insulin and growth factor signaling. When AKT phosphorylates it, FOXO1 is kept out of the nucleus; otherwise it activates genes for gluconeogenesis, stress resistance, cell cycle arrest and apoptosis. FOXO1 is studied in metabolism, diabetes, immunity and cancer.

Validated Applications

Western blot 0.1–0.5 µg/mL (Rat, Human)

Samples with a confirmed band (WB): rat lung tissue, rat brain tissue, human COLO320 cells, human HepG2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human FOXO1A fragment (Asn456–Gly655), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 93%.

Reactivity Notes

The supplier lists reactivity with human and rat. UniProt places forkhead box protein O1 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed in umbilical endothelial cells (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Rat, Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 69.7 kDa; observed band at about 70 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human FOXO1A fragment (Asn456–Gly655), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 93%.
