Product Overview
This rabbit polyclonal antibody targets forkhead box protein P3 (gene Foxp3) in mouse and rat samples and is validated for Western blot. The predicted molecular weight is 47.3 kDa, and the supplier reports an observed band at about 50 kDa.
FOXP3 is a forkhead transcription factor that is the key lineage regulator of regulatory T cells (Tregs), which suppress immune responses and maintain tolerance. It is the most widely used Treg marker, and loss-of-function mutations cause IPEX syndrome in humans and the scurfy phenotype in mice. FOXP3 is studied in autoimmunity, transplantation and tumor immunology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|
Samples with a confirmed band (WB): rat thymus tissue, mouse spleen tissue, mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide from the middle region of mouse foxp3. Sequence identity with the human ortholog: 94.4%.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places forkhead box protein P3 in the nucleus and cytoplasm. Tissue expression noted by UniProt (mouse): High level of expression in thymus and spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
