Cellular localization Cytoplasm. Nucleus. Shuttles between the nucleus and the cytoplasm. Translocated into the nucleus upon tyrosine phosphorylation and dimerization, in response to signaling by activated FGFR1, FGFR2, FGFR3 or FGFR4. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly present in the cytoplasm without stimuli. Upon leukemia inhibitory factor (LIF) stimulation, accumulates in the nucleus. The complex composed of BART and ARL2 plays an important role in the nuclear translocation and retention of STAT3. Identified in a complex with LYN and PAG1.

Product Overview

Clone 17F26 is a rabbit monoclonal antibody that recognizes forkhead box protein P3 (gene FOXP3) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 47.2 kDa, and the supplier reports an observed band at about 47 kDa.

FOXP3 is a forkhead transcription factor that is the key lineage regulator of regulatory T cells (Tregs), which suppress immune responses and maintain tolerance. It is the most widely used Treg marker, and loss-of-function mutations cause IPEX syndrome in humans and the scurfy phenotype in mice. FOXP3 is studied in autoimmunity, transplantation and tumor immunology.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): 293T cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human squamous carcinoma tissue, human tongue cancer tissue, human Hodgkin's lymphoma tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human FOXP3.

Reactivity Notes

The supplier lists reactivity with human. UniProt places forkhead box protein P3 in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 47.2 kDa; observed band at about 47 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human FOXP3.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
