Product Overview

This rabbit polyclonal antibody detects frizzled-4 (gene FZD4) in human, mouse and rat samples and is validated for Western blot and ICC/IF. The predicted molecular weight is 59.9 kDa, and the supplier reports an observed band at about 60 kDa.

Frizzled-4 (FZD4) is a Wnt receptor that also binds Norrin. Together with LRP5 and TSPAN12 it activates β-catenin signaling that drives blood vessel formation in the retina, and mutations in FZD4 cause familial exudative vitreoretinopathy. It is studied in vascular development and Wnt signaling.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Human)

Samples with a confirmed band (WB): human HepG2 cells, human PC-3 cells, human K562 cells, human RT4 cells, rat lung tissue, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): HepG2 cells.

Immunogen

Synthetic peptide from the N-terminal region of human frizzled 4. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places frizzled-4 in the cell membrane. Tissue expression noted by UniProt (human): Almost ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); Immunocytochemistry / Immunofluorescence 5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on feline samples?
A.Reactivity with feline samples has not been tested by the supplier. Aligning the immunogen sequence with the feline ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 59.9 kDa; observed band at about 60 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human frizzled 4. The peptide sequence is conserved in mouse and rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
