Product Overview
Clone 18P49 is a rabbit monoclonal antibody that recognizes ATP-dependent 6-phosphofructokinase, muscle type (gene PFKM) in human, mouse and rat samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 85.2 kDa, and the supplier reports an observed band at about 81 kDa.
PFKM is the muscle isoform of phosphofructokinase-1, which catalyzes the ATP-dependent phosphorylation of fructose 6-phosphate, the first committed step of glycolysis. It is the only PFK-1 isoform in skeletal muscle, and its inherited deficiency causes glycogen storage disease type VII (Tarui disease) with exercise intolerance.

|---|---|
| Immunocytochemistry / Immunofluorescence | 1:50–1:200 |
| Flow cytometry | 1:100 |
Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human SH-SY5Y cells, rat heart tissue, rat brain tissue, mouse heart tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human fructose 6 Phosphate Kinase.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ATP-dependent 6-phosphofructokinase, muscle type in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
