Rabbit Monoclonal

Product Overview

Clone 25F02 is a rabbit monoclonal antibody that recognizes RNA-binding protein FXR1 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 69.7 kDa, and the supplier reports an observed band at about 70–80 kDa.

FXR1 is an RNA-binding protein of the fragile X protein family, related to FMRP and FXR2. It binds target mRNAs, including AU-rich elements in 3′-UTRs, to regulate their translation and stability. FXR1 is highly expressed in muscle and testis and is studied in muscle development, spermatogenesis, neural function and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human FXR1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places RNA-binding protein FXR1 in the cytoplasm, cell projection, nucleus envelope and postsynapse. Tissue expression noted by UniProt (human): Expressed in all tissues examined including heart, brain, kidney and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 69.7 kDa; observed bands at about 70 and 80 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human FXR1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
