Product Overview

Clone 20F94 is a rabbit monoclonal antibody that recognizes FYN-binding protein 1 (gene FYB1) in human and mouse samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 85.4 kDa, and the supplier reports an observed band at about 120 kDa.

FYB1 (also called ADAP or SLAP-130) is an adaptor protein in T cells and platelets that links antigen receptor signaling to integrin activation. Working with SLP-76 (LCP2) and SKAP1, it supports T-cell adhesion and cytoskeletal remodeling, and it also contributes to platelet function. It is studied in immune cell signaling.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human TF-1 cells, human Jurkat cells, human HEL cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human FYB.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places FYN-binding protein 1 in the cytoplasm, nucleus and cell junction. Tissue expression noted by UniProt (human): Expressed in hematopoietic tissues such as myeloid and T-cells, spleen and thymus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 85.4 kDa; observed band at about 120 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human FYB.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
