Key elements and design rationale

  • Target: GAB1 (Prospero homeobox protein 1).
  • Antibody format: Monoclonal; clone AFEF-7; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse,Rat (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

GAB1 (protein: Lysosome-associated membrane glycoprotein 2 (Lamp2)) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Transcription factor involved in developmental processes such as cell fate determination, gene transcriptional regulation and progenitor cell regulation in a number of organs. Plays a critical role in embryonic development and functions as a key regulatory protein in neurogenesis and the development of the heart, eye lens, liver, pancreas and the lymphatic system. Involved in the regulation of the circadian rhythm. Represses: transcription of the retinoid-related orphan receptor RORG, transcriptional activator activity of RORA and RORG and the expression of RORA/G-target genes including core clock components: ARNTL/BMAL1, NPAS2 and CRY1 and metabolic genes: AVPR1A and ELOVL3. . Reported cellular localization context: Nucleus . RORG promotes its nuclear localization. . Tissue expression notes (as provided): Most actively expressed in the developing lens. Detected also in embryonic brain, lung, liver and kidney. In adult, it is more abundant in heart and liver than in brain, skeletal muscle, kidney and pancreas. .

Research relevance and current trends

  • Research context keywords from the source record include: Adapters,Apoptosis,Cancer,Cell Biology,Cell Cycle,Cell Differentiation,Cytoplasmic,Intracellular,Kinases,MAPK Pathway,Protein Phosphorylation,Ser/Thr Kinases,Signal Transduction.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate GAB1 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect GAB1 expression by Western blot in cell or tissue lysates, Localize GAB1 by immunofluorescence/immunocytochemistry in cultured cells, Quantify GAB1-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 45 kDa; calculated MW: 83203 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 45 kDa
  • Cellular localization (provided): Nucleus . RORG promotes its nuclear localization. .
  • Tissue details (provided): Most actively expressed in the developing lens. Detected also in embryonic brain, lung, liver and kidney. In adult, it is more abundant in heart and liver than in brain, skeletal muscle, kidney and pancreas. .

Will M01989 anti-GAB1 Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, M01989 anti-GAB1 Monoclonal antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
