Product Overview

This rabbit polyclonal antibody recognizes glutamate decarboxylase 2 (gene GAD2) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 65.4 kDa, and the supplier reports an observed band at about 65 kDa.

Glutamate decarboxylase 2 (GAD2, also called GAD65) makes the neurotransmitter GABA from glutamate. GAD65 is linked to synaptic vesicles and supplies GABA for release during high activity. It is used as a marker of GABAergic neurons and is a major autoantigen in type 1 diabetes and some neurological conditions.

Target Glutamate decarboxylase 2 (gene GAD2; UniProt Q05329, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG

Observed MW 65 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.1–0.5 µg/mL (Mouse, Rat)
IHC (paraffin sections) 0.5–1 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): rat kidney tissue, human intetsinal cancer tissue, human mammary cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Synthetic peptide from the N-terminal region of human GAD65. It differs from the mouse and rat sequences at one residue.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places glutamate decarboxylase 2 in the cytoplasm, cytoplasmic vesicle, presynaptic cell membrane and Golgi apparatus membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human, Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 65.4 kDa; observed band at about 65 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human GAD65. It differs from the mouse and rat sequences at one residue.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
