Product Overview
Clone 17G14 is a mouse monoclonal antibody that recognizes glyceraldehyde-3-phosphate dehydrogenase (gene GAPDH) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 36.1 kDa, and the supplier reports an observed band at about 36 kDa.
GAPDH catalyzes a central step of glycolysis, the conversion of glyceraldehyde-3-phosphate to 1,3-bisphosphoglycerate. Because it is abundant and broadly expressed, it is one of the most widely used loading controls for Western blot, although it also has non-glycolytic roles in the nucleus, in membrane trafficking and in translational control.

Samples with a confirmed band (WB): human HeLa cells, human Jurkat cells, human THP-1 cells, human A549 cells, rat kidney tissue, rat brain tissue, mouse kidney tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): HeLa cells.
Immunogen
Synthetic peptide based on the sequence of human GAPDH Mouse Monoclonal.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places glyceraldehyde-3-phosphate dehydrogenase in the cytoplasm, nucleus and membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
