Product Overview

This rabbit polyclonal antibody detects growth arrest-specific protein 2 (gene GAS2) in human, mouse and rat samples and is validated for Western blot and flow cytometry. The predicted molecular weight is 34.9 kDa, and the supplier reports an observed band at about 35 kDa.

Growth arrest-specific 2 (GAS2) binds actin and microtubules and is cleaved by caspases during apoptosis, which promotes cell shape changes. In the inner ear it helps maintain microtubule bundles in supporting cells, and loss of GAS2 is linked to hearing loss.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)

Samples with a confirmed band (WB): human Jurkat cells, human HepG2 cells, rat liver tissue, mouse kidney tissue, mouse liver tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): Jurkat cells.

Immunogen

Synthetic peptide from the N-terminal region of human GAS2. The peptide sequence is conserved in mouse.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places growth arrest-specific protein 2 in the cytoplasm and membrane. Tissue expression noted by UniProt (human): Ubiquitously expressed with highest levels in liver, lung, and kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 34.9 kDa; observed band at about 35 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human GAS2. The peptide sequence is conserved in mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
