Immunogen A synthesized peptide derived from human GCET2 Involved in the negative regulation of lymphocyte motility. It mediates the migration-inhibitory effects of IL6. Serves as a positive regulator of the RhoA signaling pathway. Enhancement of RhoA activation results in inhibition of lymphocyte and lymphoma cell motility by activation of its downstream effector ROCK. Is a regulator of B-cell receptor signaling, that acts through SYK kinase activation.

Key elements and design rationale

  • Target: GCSAM (Histone H3.1).
  • Antibody format: Monoclonal; clone AFHF-7; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

GCSAM (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Reported cellular localization context: Nucleus. Chromosome. Tissue expression notes (as provided): Highly expressed in the brain, with higher expression during development than in adult. Expressed also in mammary glands, testis, colon and kidney. .

Research relevance and current trends

  • Research context keywords from the source record include: Adaptive Immunity,B Cells,Cancer,Immunology,Non-CD,Tumor Biomarkers.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.

Workflow ideas (metafield): Validate GCSAM antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect GCSAM in FFPE tissue sections by immunohistochemistry, Localize GCSAM by immunofluorescence/immunocytochemistry in cultured cells, Quantify GCSAM-positive cells by flow cytometry in single-cell suspensions

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 58 kDa; calculated MW: 15404 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 58 kDa
  • Cellular localization (provided): Nucleus. Chromosome.
  • Tissue details (provided): Highly expressed in the brain, with higher expression during development than in adult. Expressed also in mammary glands, testis, colon and kidney. .

Will M12995 anti-GCET2 Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
As indicated on the product datasheet, M12995 anti-GCET2 Monoclonal antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
