Rabbit Polyclonal

Product Overview

This rabbit polyclonal antibody detects zinc finger protein Gfi-1 (gene GFI1) in human, mouse and rat samples and is validated for IHC. The predicted molecular weight is 45.3 kDa.

GFI1 (growth factor independence 1) is a zinc-finger transcriptional repressor that recruits chromatin modifiers such as LSD1 (KDM1A) and HDACs. It is essential for neutrophil development and for hematopoietic stem cell quiescence, and it also acts in lymphocytes and inner ear hair cells. GFI1 mutations cause a form of severe congenital neutropenia.

Target Zinc finger protein Gfi-1 (gene GFI1; UniProt Q99684, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human, Mouse, Rat
Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4; 0.05 mg NaN3
Calculated MW 45.3 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL

Samples with confirmed staining (IHC): human mammary cancer tissue, human rectal cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).

Immunogen

Synthetic peptide from the C-terminal region of human GFI1. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places zinc finger protein Gfi-1 in the nucleus.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: IHC (paraffin sections) 0.5–1 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on primate samples?
A.Reactivity with primate samples has not been tested by the supplier. Aligning the immunogen sequence with the primate ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of human GFI1. The peptide sequence is conserved in mouse and rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
