Vendor description (summary): Boster Bio Anti-GITR/Tnfrsf18 Picoband® Antibody catalog # A03125-3.

Biological background

Biological context: Receptor for TNFSF18. Seems to be involved in interactions between activated T-lymphocytes and endothelial cells and in the regulation of T-cell receptor-mediated cell death. Mediated NF-kappa-B activation via the TRAF2/NIK pathway.

Expression and localization notes: cellular localization: Cell membrane. Single-pass type I membrane protein. Secreted., tissue context: Detected in thymus and in activated splenocytes..

Common research applications

  • Western blotting (WB): Compare Tnfrsf18 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of Tnfrsf18 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify Tnfrsf18-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: Tumor necrosis factor receptor superfamily member 18 (TNFRSF18) also known as activation-inducible TNFR family receptor (AITR) or glucocorticoid-induced TNFR-related protein (GITR) is a protein that in humans is encoded by the TNFRSF18 gene. It is mapped to 4; 4 E2. This gene encodes a member of the TNF-receptor superfamily. The encoded receptor has been shown to have increased expression upon T-cell activation, and it is thought to play a key role in dominant immunological self-tolerance maintained by CD25 (+)CD4 (+) regulatory T cells. Knockout studies in mice also suggest the role of this receptor is in the regulation of CD3-driven T-cell activation and programmed cell death. Three alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane. Single-pass type I membrane protein. Secreted.
  • Tissue details: Detected in thymus and in activated splenocytes.
  • Research category: Adaptive Immunity,Cancer,Cytokines,Growth Factors,Growth Factors/Hormones,Immunology,Innate Immunity,Signal Transduction
