Product Overview

This rabbit polyclonal antibody recognizes gap junction beta-2 protein (gene GJB2) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 26.2 kDa, and the supplier reports an observed band at about 26 kDa.

Connexin 26 (GJB2) is a gap junction protein that forms channels linking the cytoplasm of adjacent cells. In the cochlea it supports ion recycling needed for hearing, and GJB2 mutations are the most common genetic cause of inherited non-syndromic hearing loss, so it is widely studied in auditory research.

Target Gap junction beta-2 protein (gene GJB2; UniProt P29033, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG
Reactivity Human, Mouse, Rat
Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4
Calculated MW 26.2 kDa
Observed MW 26 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.25–0.5 µg/mL (Mouse, Rat)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): rat liver tissue, rat kidney tissue, mouse liver tissue, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human GJB2 fragment (Ala88–Thr137), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places gap junction beta-2 protein in the cell membrane and cell junction. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Mouse, Rat); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 26.2 kDa; observed band at about 26 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human GJB2 fragment (Ala88–Thr137), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
