Product Overview
Clone AOAF-7 is a rabbit monoclonal antibody that recognizes pro-glucagon (gene GCG) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 20.9 kDa, and the supplier reports an observed band at about 21 kDa.
Proglucagon (GCG) is processed into several hormones depending on the tissue. In pancreatic α cells it yields glucagon, which raises blood glucose by stimulating hepatic glucose output, and in intestinal L cells it yields GLP-1 and GLP-2, which boost insulin secretion and support gut growth. GCG products are central to studies of glucose homeostasis, diabetes and obesity.

| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): HepG2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Samples with confirmed staining (IHC): mouse pancrease tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human glucagon.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes pro-glucagon as a secreted protein. Tissue expression noted by UniProt (human): [Glucagon]: Secreted in the A cells of the islets of Langerhans... The samples tested by the supplier (listed above) are a practical starting point for positive controls.
