Product Overview
This rabbit polyclonal antibody recognizes glucose-6-phosphate isomerase (gene GPI) in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 63.1 kDa, and the supplier reports an observed band at about 63 kDa.
Glucose-6-phosphate isomerase (GPI) interconverts glucose 6-phosphate and fructose 6-phosphate, the second step of glycolysis and a reverse step in gluconeogenesis. Outside the cell the same protein acts as a cytokine known as autocrine motility factor or neuroleukin, and GPI deficiency causes hemolytic anemia.

Samples with a confirmed band (WB): human SiHa cells, human A549 cells, human K562 cells, human A431 cells, rat lung tissue, rat heart tissue, mouse lung tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): K562 cells.

Synthetic peptide from the N-terminal region of human GPI. It differs from the mouse and rat sequences at sixteen residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes glucose-6-phosphate isomerase as secreted and also located in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
