Product Overview

Clone 7B6 is a mouse monoclonal antibody that recognizes guanine nucleotide-binding protein G(I)/G(S)/G(O) subunit gamma-4 (gene GNG4) in mouse and rat samples and is validated for Western blot. The predicted molecular weight is 8.4 kDa, and the supplier reports an observed band at about 12 kDa.

Gγ4 (GNG4) is a gamma subunit of heterotrimeric G proteins. Bound to a Gβ subunit, it forms the Gβγ dimer that is required for G protein activation and relays signals from G protein-coupled receptors to downstream effectors; it is expressed notably in the brain.

Validated Applications

Western blot 0.25–0.5 µg/mL (Mouse, Rat)

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Recombinant human GNG4 fragment (Met1–Asp52), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt places guanine nucleotide-binding protein G(I)/G(S)/G(O) subunit gamma-4 in the cell membrane. Tissue expression noted by UniProt (human): Brain, kidney, pancreas, skeletal muscle and faintly in cardiac muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 8.4 kDa; observed band at about 12 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human GNG4 fragment (Met1–Asp52), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
