Product Overview
This rabbit polyclonal antibody detects growth factor receptor-bound protein 14 (gene GRB14) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 61.0 kDa, and the supplier reports an observed band at about 61 kDa.
GRB14 is an adaptor protein of the GRB7/10/14 family that binds the activated insulin receptor. It inhibits the receptor's kinase activity and down-stream MAPK signaling, while also helping recruit PDK1 to the membrane, so it is studied in insulin action and metabolic regulation.

Samples with a confirmed band (WB): human A549 cells, human HEK293 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human renal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with a confirmed signal (flow cytometry): U87 cells.

Reactivity Notes
The supplier lists reactivity with human. UniProt places growth factor receptor-bound protein 14 in the cytoplasm and endosome membrane. Tissue expression noted by UniProt (human): Expressed at high levels in the liver, kidney, pancreas, testis, ovary, heart and skeletal muscle. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
