Product Overview
This rabbit polyclonal antibody targets G protein-coupled receptor kinase 5 (gene GRK5) in human, mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 67.8 kDa, and the supplier reports an observed band at about 68 kDa.
GRK5 is a membrane-associated G protein-coupled receptor kinase that phosphorylates activated adrenergic, muscarinic, dopamine and opioid receptors, leading to β-arrestin binding, desensitization and internalization. Unlike most GRKs, it can also enter the nucleus and act on non-receptor targets. GRK5 is studied in cardiac hypertrophy, inflammation and GPCR signal regulation.

Validated Applications
| Western blot | 0.1–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL (Human, Mouse, Rat) |
Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human A549 cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat cardiac muscle tissue, mouse lung tissue, human placenta tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of human GRK5. It differs from the mouse and rat sequences at three residues.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places G protein-coupled receptor kinase 5 in the cytoplasm, nucleus and cell membrane. Tissue expression noted by UniProt (human): Highest levels in heart, placenta, lung > skeletal muscle > brain, liver, pancreas > kidney. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
