Product Overview
This rabbit polyclonal antibody targets endoplasmic reticulum chaperone BiP (gene HSPA5) in rat, mouse and human samples and is validated for Western blot and IHC. The predicted molecular weight is 72.3 kDa, and the supplier reports an observed band at about 78 kDa.
BiP (HSPA5, also called GRP78) is the main HSP70 chaperone of the endoplasmic reticulum lumen. It helps fold and assemble newly made secreted and membrane proteins, holds misfolded ones for degradation, and controls the unfolded protein response sensors IRE1, PERK and ATF6. It is a standard marker of ER stress.

Validated Applications
| Western blot | 0.025–0.25 µg/mL (Human, Mouse, Rat) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
Samples with a confirmed band (WB): human U251 cells, human HepG2 cells, human K562 cells, human A431 cells, rat brain tissue, rat liver tissue, mouse brain tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.17 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human cerebral cortex tissue, human liver cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of human GRP78 BiP. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with rat, mouse and human. UniProt places endoplasmic reticulum chaperone BiP in the endoplasmic reticulum lumen, melanosome, cytoplasm and cell surface. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
