Product Overview
This rabbit polyclonal antibody recognizes hematopoietic lineage cell-specific protein (gene HCLS1) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 54.0 kDa, and the supplier reports an observed band at about 75 kDa.
Hematopoietic lineage cell-specific protein (HCLS1, HS1) is an actin-binding protein expressed in blood cells and related to cortactin. It is phosphorylated after antigen receptor engagement and helps organize the actin cytoskeleton at the immune synapse and during cell migration. HS1 is studied in lymphocyte signaling and B-cell leukemia.

Samples with a confirmed band (WB): human Jurkat cells, human Daudi cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): RT4 cells.
Immunogen
Recombinant human HCLS1 fragment (Asn213–Lys473), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places hematopoietic lineage cell-specific protein in the membrane, cytoplasm and mitochondrion. Tissue expression noted by UniProt (human): Expressed only in tissues and cells of hematopoietic origin. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
