Anti-HCN2 Antibody (#APC-030) is a highly specific antibody directed against an epitope of the human protein. The antibody can be used in western blot, immunoprecipitation, immunohistochemistry, and immunocytochemistry applications. It has been designed to recognize HCN2 from human, rat, and mouse samples. Anti-HCN2-ATTO Fluor-594 Antibody (#APC-030-AR) is directly labeled with ATTO-594 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-594 fluorescent label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594. The conjugated antibody is ideal for multiplex staining studies.

Target HCN2

Overview

Anti-HCN2-ATTO Fluor-594 Antibody is an antibody targeting Hyperpolarization-activated cyclic nucleotide-gated potassium channel 2, BCNG2, HAC-1 Polyclonal raised in Rabbit (ATTO-594. Maximum absorption 601 nm; Maximum fluorescence 627 nm. The fluorescence is excited most efficiently in the 580 - 615 nm range. This label belongs to the class of Rhodamine dyes and can be used with fluorescent equipment typically optimized to detect Texas Red and Alexa-594.). This antibody is commonly used in IF, IHC to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Hyperpolarization-activated cyclic nucleotide-gated potassium channel 2, BCNG2, HAC-1 (also reported as Hyperpolarization-activated cyclic nucleotide-gated potassium channel 2, BCNG2, HAC-1).
  • Immunogen/epitope region: Intracellular, N-terminus.
  • Homology note: Rat, mouse - identical (informative for cross-species interpretation).
  • Species reactivity (as provided): Human, Rat, Mouse.
  • KO-validated: yes (validation context may be assay-dependent).
  • Cited use: IP, IHC, ICC (literature use does not guarantee performance in every setup).
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #APC-030), and immunohistochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

Hyperpolarization-activated cation currents (Ih) appear in the heart and the brain having crucial role in controlling electrical pacemaker activity, contributing to biological processes such as heartbeat, sleep-wake cycle and synaptic plasticity.1,2The Ih currents are generated by the hyperpolarization-activated cyclic nucleotide-gated channel family (HCN), which comprises four homologous members, named HCN1-4. Each HCN subunit consist of six transmembrane domains (TM), a pore region between TM5-TM6 and a binding domain to cyclic nucleotides (CNBD) in the cytoplasmic C-terminus.2The HCN subunits can form functional homomers and can also co-assemble into functional heteromers.2The channels are closely related to each other and share homology of about 60%. However, they are diverging from each other in their cytoplasmic N- and the C-terminus.

Research relevance and current trends

  • Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
  • Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
  • Combining antibody readouts with functional assays for more complete interpretation.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Conceptual control: KO/KD samples provide orthogonal support for target assignment when available.
  • Provided control suggestions: Negative control: BLP-PC030.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
