Product Overview
This rabbit polyclonal antibody detects potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 (gene HCN4) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 129.0 kDa, and the supplier reports an observed band at about 140 kDa.
HCN4 is the main hyperpolarization-activated cyclic nucleotide-gated channel of the sinoatrial node, where it carries the funny current (If) that sets heart rate. It activates slowly on hyperpolarization and is enhanced by cAMP, linking heart rate to sympathetic input. HCN4 variants cause sinus node dysfunction, and it is the target of the heart-rate-lowering drug ivabradine.

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Recombinant human HCN4 fragment (His909–Leu1203), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places potassium/sodium hyperpolarization-activated cyclic nucleotide-gated channel 4 in the cell membrane. Tissue expression noted by UniProt (human): Highly expressed in thalamus, testis and in heart, both in ventricle and atrium. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
