Product Overview

This rabbit polyclonal antibody targets hypocretin neuropeptide precursor (gene HCRT) in mouse and rat samples and is validated for IHC and ICC/IF. The predicted molecular weight is 13.4 kDa.

The hypocretin precursor (HCRT) is processed into two neuropeptides, orexin-A and orexin-B, made by a small group of neurons in the lateral hypothalamus. Acting on the OX1R and OX2R receptors, they promote wakefulness and regulate feeding and energy balance. Loss of orexin neurons causes narcolepsy with cataplexy, and HCRT is studied in sleep and metabolism.

Target Hypocretin neuropeptide precursor (gene HCRT; UniProt O43612, human)
Host / Clonality / Isotype Rabbit / Polyclonal / IgG
Reactivity Mouse, Rat
Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4
Calculated MW 13.4 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

IHC (paraffin sections) 2–5 µg/mL (Mouse, Rat)
Immunocytochemistry / Immunofluorescence 5 µg/mL (Mouse, Rat)

Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue, rat cerebellum tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Samples with confirmed staining (ICC/IF): mouse brain tissue, rat brain tissue.

Immunogen

Synthetic peptide from the middle region of human HCRT. The peptide sequence is conserved in mouse and rat.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt places hypocretin neuropeptide precursor in the rough endoplasmic reticulum, cytoplasmic vesicle and synapse. Tissue expression noted by UniProt (human): Abundantly expressed in subthalamic nucleus but undetectable in other brain regions tested (hypothalamus was not tested) and in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: IHC (paraffin sections) 2–5 µg/mL (Mouse, Rat); Immunocytochemistry / Immunofluorescence 5 µg/mL (Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human HCRT. The peptide sequence is conserved in mouse and rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
