Product Overview
This rabbit polyclonal antibody recognizes histone deacetylase 1 (gene HDAC1) in human samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 55.1 kDa, and the supplier reports an observed band at about 65 kDa.
Histone deacetylase 1 (HDAC1) is a class I deacetylase that removes acetyl groups from lysines on core histones and other proteins. Removing acetyl groups from histone tails generally compacts chromatin and represses transcription. It works within corepressor complexes such as Sin3, NuRD and CoREST, often with HDAC2, and is studied in gene regulation, development and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL/ml (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL/ml (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/mL/1×106 cells (Human) |
Samples with a confirmed band (WB): human MOLT-4 cells, human Jurkat cells, human HL-60 cells, human U251 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human esophageal squamous carcinoma tissue, human lung cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): MCF-7 cells.
Immunogen
Synthetic peptide from the middle region of human HDAC1. The peptide sequence is conserved in mouse and rat.
Reactivity Notes
The supplier lists reactivity with human. UniProt places histone deacetylase 1 in the nucleus. Tissue expression noted by UniProt (human): Ubiquitous, with higher levels in heart, pancreas and testis, and lower levels in kidney and brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
